Studies on Hepatic Microsomal Cytochrome P-450 Related Detoxifying Enzyme System in Indigenous Korean Cattle and Yorkshire Pig


Abstract
The concentration and activity of cytochrome P-450 related drug metabolizing enzymes(NADPH-cytochrome creductase, benzpyrene hydroxylase in the hepatic microsomes prepared from slaughtered Yorkshire pig and Indigenous Korean cattle in Chonju area were assayed.
The absorption spectra of CO-binding cytochrome P-450 in the hepatic microsomes from cattle and pig showed the typical peak at wavelength 450mm respectively as showed at rats. The cytochrome P-450 concentration of the hepatic microsomes from swine were higher as 0.59" }0.10 nmol"^mg protein in male than 0.46" }0.12 in female. The activity of the NADPH-cytochrome c reductase, the component of cytochrome P-450 system and the drug metabolizing enzymes showed the same tendency as the cytochrome P-450 concentration in both gender of pig. The cytochrome P-450 concentration in the hepatic microsomes in Indigenous Korean cattle were higher as 0.88" }0.11 nmol"^mg protein in male than 0.73" }0.13 in female. The activity of the hepatic microsomal cytochrome P-450 related drug metabolizing system in bovine showed the same tendency as the differences in both gender of pig, but the metabolizing capacity on the same drug was significantly affected by species as benzpyrene hydroxylase activity in cattle was 5 times higher than that in pig.
